A decrease in the tiller number and male sterility will lead to a decline rice yield. Therefore, it is significant study molecular mechanism of controlling regulating reproductive development. The mutant st2 (single 2) was induced by ethyl methane sulfonate (EMS) indica maintainer line Xinong 1B showed single tillering sterility. I2-KI staining that pollen aborted. scanning electron microscope (SEM) observation underlined anther became smaller, wax epidermis reduced, inner wall shrank Ubisch body decreased, collapsed, germination pore developed abnormally. genetic analysis discovered trait controlled recessive nuclear gene located on chromosome 3. LOC_Os03g05540 encoding tetratricopeptide repeat (TPR) domain identified as candidate sequencing. quantitative real-time polymerase chain reaction (qRT-PCR) indicated ST2 highly expressed stem apical meristem (SAM) initial stage meiosis during subcellular localisation plasmic protein. homology demonstrated evolutionarily conserved. These results laid foundation for further function.
